時期特異的・部位特異的遺伝子破壊法を用いたShn2遺伝子の行動学的機能解析

Research Abstract

本研究は、顕著な精神疾患様の行動異常を示し、非常に妥当性の高い精神疾患のモデルマウスと考えられるSchnurri-2欠損マウス(Shn2 KOマウス)について、脳のどの部分におけるどの時期のShn2の欠損がこれらの多様な行動学的異常をもたらしているのかを知るために、時期特異的・領域特異的Shn2欠損マウスを用いて行動を解析することを目的としている。
この目的のため、Shn2のexon領域の配列をloxP配列で挟んだShn2 floxedマウスを入手した。そして各部位・各時期に特異的にCreリコンビナーゼを発現させるCREマウスと交配させるために十分な数を繁殖させた。また、Shn2 floxedマウスにおいてloxP配列が機能するかどうかを確認するたあに、CaMKIIプロモーター支配下にCreを発現させるCK-CREマウスと交配させ、(CK-CRE/W ; floxed/w)という遺伝子型のマウスを得た。さらにこのマウスに(floxed/w)の遺伝子型のマウスを交配させることで(CK-CRE/w ; floxed/floxed)という遺伝子型のマウスを得ることができた。このマウスではCREが働いた細胞でShn2が欠失しているはずであり、誕生したマウスの尻尾を用いて遺伝子型の判定をするとShn2が欠失している個体が見られた。本来、CaMKII CREマウスは前脳特異的に働いており、尻尾では組換えは起こらないはずであるが、発生のどこかの段階でCaMKIIか発現し、組換えが起こったものと考えられる。Shn2 floxedマウスにおいてCREによる組換えが起こることは確認できたが、時期特異的・領域特異的に組換えを起こさせるためにはCREマウスの選択や交配の組み合わせを注意する必要がある。
