生細胞導入DNAビーズを使った小胞体・核膜様の膜のアセンブリー機構の解明

| Research Abstract |
本研究は、生細胞内に導入したDNAビーズの周囲に観察される"非分解系"の膜構造について、(1)膜の集合機構を顕微鏡イメージングによって解明し、(2)ビーズの初期認識に関わる細胞内センサー因子を同定することで、細胞による外来異物の認識機構の一端を解明することを目的としている。まず、エンドサイトーシスによってDNAビーズをHeLa細胞内に取り込ませ、各種オルガネラマーカータンパク質に対する抗体を用いて免疫染色を行ったところ、DNAビーズの周囲に、核膜内膜タンパク質であるemerinをはじめとするLEMドメインタンパク質、および、LEMドメインタンパク質への結合能を有するbarrier-to-autointegration factor (BAF)が局在していることが分かった。次に、EGFP-BAFを安定発現するHeLa細胞にDNAビーズを導入し、BAF陽性となってからの経過時間毎にサンプルを固定してビーズ周囲に存在する膜構造を電子顕微鏡で観察したところ、BAF陽性となってから約10分が経過した時点で、すでにDNAビーズ周囲を取り囲むような連続した膜構造が観察された。BAFには、in vitroでDNA配列非依存的にDNAと結合する機能があることから、本実験結果は、BAFが細胞内でもDNAビーズに結合し、LEMドメインタンパク質を含む膜をビーズ周囲に集合させる働きを持つことを示している。以上のデータは、BAFが外来DNAを認識する細胞内センサーとして働いている可能性を示唆している。
