Research Abstract |
This study aimed to explore a novel bioactive molecule that regulate ion channel function by in silico screening with a pharmacophore model of its chemical ligands. Sphingosine fits the pharmacophore model for inwardly rectifier potassium(Kir) channel 4. 1 blockers. Sphingosine has an inhibitory effect on Kir4. 1 channels. When biosynthesis of sphingosine from sphingomyeline via ceramide is stimulated, Kir4. 1 channel currents decrease. These results suggest that endogenous sphingolipids inhibit Kir4. 1 channels via sphingosine.
