HIV潜伏感染モデル動物の構築と潜伏化メカニズムの解明

Research Abstract

Human immunodeficiency virus (HIV)を感染者体内から完全除去する事は、現在の薬剤療法では不可能である。HIVの潜伏感染がその原因であると考えられているが、その潜伏化機構はいまだに明らかになっていない。そこで本研究では、HIV潜伏感染決定メカニズムの解明を目的とした。
H22年度は、生体内におけるHIVの潜伏感染を再現するために、LTRプロモーターよりウイルスの転写調節因子TatとGFPの発現のみを行う非増殖型HIVベクター(singl eround-HIV, SR-HIV)を用いて、ヒト血液幹細胞移植マウス(ヒト化マウス)で感染実験を行った。SR-HIV感染2週間後のヒト化マウスの抹消Tリンパ球において、恒常的GFP陽性細胞と、CD3とIL-2の増殖刺激環境下でGFP発現が誘導される細胞が検出された。しかしながら、分離解析できるほどの感染細胞数は得られなかった。さらなる解析のためにはSR-HIVのヒト血液幹細胞への導入効率を増加させる手法を用いる必要がある。
H23年度はHIVプロウイルスの組込み位置とHIVの潜伏化の相関を解析することを目的として、linker PCR法を用いたHIVの組込み位置解析方法を確立した。これまでの我々の研究で、インテグラーゼ(IN)非依存的に組込まれたHIVプロウイルスからのウイルス発現量は、IN依存的に組込まれたものに比して低い事が明らかになっていた。H23年度はこの方法を用いて、ウイルス組込み経路の違いにより誘導されるウイルス発現量の違いが、HIVの組込み位置によって決定されていることを明らかにした(Virology. 2012 25;427(1):44-50)。
