Outline of Annual Research Achievements

本研究課題のアプローチの主軸である、1次元の収差補正と複数方向の走査データの組わせによる分解能向上について、in vivo ヒト皮膚でのデジタルフォーカス補正による原理検証を行った。狭い走査範囲でのモーション補正と、一次元デジタルフォーカス補正と複数方向の走査データを組み合わせた補正をかけることにより、不随意運動による位置ずれが補正され、デフォーカスによる皮膚内部構造のボケが改善された。ピント位置(インフォーカス)から遠い深さ位置でも微細な内部構造を可視化できることが確認できた。
また、高次収差を含めた 3次元 OCT データを補正するための理論モデルを構築し、基本アルゴリズムの実装を行った。微粒子試料を用いた検証により、高次収差が存在する場合でも、3次元 OCT データの補正を各深さ毎に処理することなく達成できることが確認できた。ただし、高次収差を含む場合、補正パラメータの推定は多次元最適化問題となり、収束に難がある場合が確認された。
In vivo ヒト網膜イメージングにおいて、プローブ光を Lissajous パターンで走査しつつ走査位置をずらすことで、高密度空間サンプリングと広い画角を達成した [Makita et al., Biomed. Opt. Express 13, 5212-5230 (2022)]。この手法と本課題で提案する収差補正方法の組合わせることで、広い画角で収差補正した高分解能網膜 OCT イメージングを実現することが期待される。

Reason

第一の目的である1次元補正と複数方向の走査データを組み合わせる本アプローチの検証は、デフォーカスのみの基礎的な動作検証は達成された。今後は、より高次の収差を含んでいる場合への適応を検証していく。
高次収差を含む場合の補正方法も、試料を用いた検証により動作確認が行われた。
これらのことから、高次収差を含む場合の in vivo 生体高分解能 OCT イメージングへの事前準備がスムーズに進行しているため、本課題は当初の計画通りに順調に進展していると考えられる。

Strategy for Future Research Activity

In vivo 生体での高次収差も補正するアプローチの動作検証と問題点の洗い出し・解決案の検討と実装を行う。
上記概要でも触れた、多次元最適化問題へのアプローチを検討し、最適化アルゴリズム・最適化コスト関数の見直しと組合せの検討をする。
これら進めていき、in vivo 生体での三次元高分解能イメージングの実現を目指す。

Causes of Carryover

当初、初年度で購入予定であった高速計算処理用の計算機の購入を次年度に変更したため、次年度使用額が生じた。現行の計算機で計算処理を行うことで、要求されれる計算量、データ量を見積もることができたため、より適した計算機の購入を検討することが可能となった。
