Outline of Annual Research Achievements

Heart disease is associated with various organs, including the intestines, kidneys, and bones. Osteocrin (OSTN) and osteoprotegerin (OPG), which play an important role in bone metabolism, and associate with myxomatous mitral valve disease (MMVD) and various cardiac tests in humans; however, this remains unclear in the veterinary field. This study evaluated the association of OSTN and OPG with MMVD severity and various cardiac tests in dogs. We included healthy dogs (H) and dogs with MMVD, who were grouped based on the ACVIM classification (stage B1, B2, C/D) based on cardiac murmur intensity, echocardiography, and thoracic radiography findings. Serum OSTN and OPG levels were measured, and their relationship with various parameters was evaluated. Among dogs with MMVD, serum OSTN levels were significantly lower in group B1 than in group H; in group B1 than in group B2; and in groups H, B1, and B2 than in group C/D. Positive correlations were also found between serum OSTN levels and left atrium, left atrium to aorta ratio, left ventricular internal diameter at diastole, left ventricular inflow E wave, and left ventricular fractional shortening. In contrast, serum OPG levels were markedly higher in group B2 than in group B1; however, there were no significant differences among the other groups. Our findings indicate a relationship between heart disease and bone metabolism and that in, dogs with MMVD, serum OSTN, but not OPG, levels may be positively correlated with MMVD severity.

大学業務が昨年度過多になったため、研究に要する時間確保が困難であった。
現在、犬の心臓病と骨バイオマーカーの関連性は投稿中であるが、続く、ラット心不全モデルでの心骨連関の研究を遂行中である。

Strategy for Future Research Activity

現在、ラットの心不全モデル作製前であるが、モデル作製後1か月で骨を採材し、外部の検査会社にて骨強度、骨密度、骨の組織の検討予定である。また、血清中のNT-proBNP、オステオプロテゲリン、オステオクリンの濃度をELISAで測定する予定であるが、既に各ELISA kitは購入中のため、今年の夏ごろまでには研究が終了し、骨と心臓の関連性の基礎的検討が終了し、論文化をする予定である。
