Outline of Final Research Achievements

Children with short bowel syndrome (SBS) develop intestinal failure-associated liver disease (IFALD) at a high rate under fasting and total parenteral nutrition (TPN) management. IFALD is an important pathology that determines life prognosis. On the other hand, adrenomedullin (AM) has a variety of physiological activities and is expected to have a protective effect on multiple organs. In this study, we aimed to invent a treatment method that overcomes SBS and IFALD simultaneously by administering AM to SBS/IFALD model rats, thereby regenerating the intestine and improving liver damage.
As a preliminary step to administering adrenomedullin, hepatocyte growth factor and clodronate, which are thought to have a similar physiological activity, were administered, and these prevented or improved IFALD, which is mainly caused by fatty liver.

Academic Significance and Societal Importance of the Research Achievements

短腸症候群患児らにとって生命予後に直結するIFALDは、未だその治療法は確立されておらず、また肝障害の多くは不可逆的とされているので、IFALDの予防または治療法の開発は喫緊の課題である。本研究成果は、IFALD克服に向けた成果となり得る。
