Outline of Final Research Achievements

Among the complications of chronic kidney disease (CKD), the development of cardiovascular disease in particular is closely related to life expectancy. We found that vitamin A is associated with abnormal immunoglobulin A (IgA) secretion and accumulation of uremic substances in CKD mice. We analyzed intestinal IgA-producing cells and found that retinoic acid secretion by dendritic cells in Peyer's patches is enhanced in CKD mice, which promotes the germinal center response, a differentiation response to IgA-producing cells.

Academic Significance and Societal Importance of the Research Achievements

本研究における一連の解析によって、CKD 病態における腸内細菌治療の意義と新たな治療標的が示された。本研究のさらなる解析が腸内細菌由来尿毒症物質の蓄積抑制を目的とした新規治療薬の開発に繋がることが期待される。
また、上流の原因物質であることを見出したビタミンAは、現状の人工血液透析法では血中からの除去が困難であるため、血液透析の技術発展が望まれる。また、得られた病態関連分子を標的とした新たなCKD治療法の開発が期待される。
