Outline of Final Research Achievements

A group of immune guidance molecules have distinct effects on extracellular trap formation and contribute to neutrophil- and eosinophil-associated diseases. We reported that integrin-mediated adhesion contributes to extracellular trap formation in vasculitis (Immunohorizons 2022) and that mitochondrial DNA contributes to neutrophil activation in Behcet's disease (EMBO J 2023). We also proposed a prognostic prediction based on immunological profiles of peripheral blood mononuclear cells from patients with vasculitis by subjecting them to single-cell analysis (Nat Commun 2023).

Academic Significance and Societal Importance of the Research Achievements

本邦において厚生労働省指定難病に該当する顕微鏡的多発血管炎やベーチェット病について好中球活性化のメカニズムの一端を明らかにし、新たな治療戦略につながることが期待される。また、シングルセル解析を介して、好中球のみならず単球の遺伝子発現プロファイルに基づいた層別化に成功し、疾患予後予測へつながる成果を見いだした。
