Outline of Final Research Achievements |
For the further analysis about uterine dendritic cells (DCs) for induction of feto-maternal tolerance, DC subset analysis was performed. The IDO-positive cells induce immune tolerance through production of IL-10. Although PD-L2+ DCs were detected by analysis of uterus DCs of allogeneic pregnancy mouse Day3.5, those were not expressed IDO. This examination did not lead to the expansion of choice as the another tolerogenic DC subset. We reported the dynamic changes of uterine DCs during pre-implantation. Additionally we performed more detailed analysis of regulatory cells (Tregs) during pre-implantation period. The change of Tregs during pre-implantation was similar to DCs, it was confirmed the interaction between DCs and Tregs.

Academic Significance and Societal Importance of the Research Achievements |
妊娠・出産年齢の高年齢化に伴い、不妊症・不育症の有病率は増加傾向にある。原因不明の着床不全に免疫学的機序が関わる可能性があり、その原因究明、治療薬の解明は不妊症・不育症の治療につながる可能性がある。近年、抗癌治療において、免疫チェックポイント阻害薬の適応拡大は顕著である。治療ターゲットとして、免疫機構は大いに注目されており、妊娠免疫における機構解明が求められている。妊娠中の免疫寛容の誘導における樹状細胞の役割はいまだに不明な部分が多く、その性状の詳細な解析により、免疫寛容の誘導メカニズムの一端が明らかになる可能性がある。
