Outline of Final Research Achievements

Single cell RNA-sequencing in young (8 weeks old) and old (80 weeks old or older) wild-type mice using bone marrow cells collected from each group revealed that the sinusoidal basal Adiponectin (ADIPOQ)-positive CAR cells (Adipo-CAR) localized around the membrane change with aging of bone marrow, and many of the genes whose expression changes in Adipo-CAR are related to the basement membrane. Although, immunohistological staining of the basement membrane of each group revealed no significant differences.
Next, in order to clarify which basement membrane molecules are involved in the maintenance of CAR cells, we cultured CAR cells from cxcl12-GFP mice using GFP as an indicator in culture dishes coated with laminin 111, 221, 332, 411, 511 and PBS(control). However, after 3 days of culture, no GFP signal was observed in any culture dish.

Academic Significance and Societal Importance of the Research Achievements

骨髄の血管基底膜がCAR細胞の機能維持において重要な役割を果たしていることはすでに明らかとなっているが,骨髄の血管に特異的に発現する基底膜分子や,それら基底膜分子が老化に伴いどのように変化するのか,という点については不明であった.
本研究では,老化により類洞基底膜周囲に局在するAdiponectin (ADIPOQ)陽性CAR細胞が変化し,これらのCAR細胞の発現変動遺伝子に基底膜関連のものが多く含まれることが明らかとなった.これらの発見は,加齢による免疫機能低下のメカニズムを解明し,骨髄老化の関与が報告されている歯周病やインプラント周囲炎に対する予防及び治療法の開発へ繋がるものと考えられる.
