Outline of Final Research Achievements |
In this study, we aimed to elucidate the site-specific mechanisms of inflammation control in the large and small intestines through vascular research. To date, we have created endothelial specific knockout mouse model in which the blood vessels are not maintained. Preliminary experiments have also revealed that the mechanisms controlling inflammation may differ between the small intestine and the large intestine. Using immunostaining, FACS analysis, gene expression analysis, and endothelial cell culture experiments, we analyzed the vascular structure of the intestines, the distribution of blood cells, and differences in gene expression. As a result, several genes were expressed differently, and we focused on a fraction of cytokines and immune cells to analyze the relationship between vascular maintenance mechanisms and inflammation control mechanisms. Although we could not identify the key molecule in inflammation control, we are continuing to analyze other candidates.

Academic Significance and Societal Importance of the Research Achievements |
腸炎はヒトが生活する中で、最も頻繁に経験する炎症反応の一つである。また、原因不明の炎症性腸疾患も増加の一途をたどっている。腸で生じる炎症は、腸管の部位特異性を認めることがあるが、なぜこのような部位特異性を認めるか不明である。本研究では、血管研究を通じて、大腸と小腸における部位特異的炎症制御機構を解明に挑戦した。今回、腸管の血管構造、免疫細胞の分布を明らかにして、血管維持機構との関係を、培養実験を通じて解析した。炎症制御に関与すると考えられる候補因子を複数同定しており、今後これらを制御することで、腸管の炎症制御につながることが期待できる。
