Outline of Annual Research Achievements

ゲノム情報を制御し細胞の性質を運命づけるゲノム修飾情報であるエピゲノムは、その異常により消化器癌など多くの悪性腫瘍の発生と進展に寄与している。それゆえ、癌ドライバーとして機能し、治療標的となり得るドライバーエピゲノム異常を同定し、それら重要なエピゲノムを標的とした機能性分子を開発して新たな治療基盤を創生することが、特に薬剤抵抗性の難治癌症例に対して求められている。エピゲノムに対する従来の機能性分子単剤では作用領域がゲノム全体に及ぶ弊害があるため、本研究では、DNA配列を認識する小分子ピロール-イミダゾールポリアミド(PIP)を機能性分子に融合させ、作用領域を局所化させる技術開発を行った。ヒストン脱メチル化酵素阻害剤やヒストンアセチル化酵素阻害剤の構造を改変・簡略化し、PIPと縮合してそれぞれ腫瘍細胞へ投与し増殖抑制作用を確認するとともに、PIPが認識する配列を含む領域において有意にエピゲノム改変が起きることを確認した。癌で治療標的となり得る重要なエピゲノム異常領域を同定する研究では、EBウイルス感染によるエピゲノム異常が発癌に寄与する胃癌や上咽頭癌などのEBV関連腫瘍について解析した。EBV感染によりEBVゲノムがヘテロクロマチンに結合しエンハンサーの活性化マークを上昇させ癌原遺伝子を発現上昇させる「エンハンサー侵襲」が胃癌だけでなく上咽頭癌でも重要であることを報告した。またHi-ChIP法を用いて、癌で特異的に異常形成されるエンハンサー・プロモーター間の近接関係を網羅的に解析した。それら異常活性化エンハンサーに有意に認める転写因子結合モチーフ配列を同定し、候補群の中からTCF7が重要な転写因子であり、下流標的として癌遺伝子MYBの異常活性化が癌細胞増殖に重要であること、CRISPRiによるエンハンサーの特異的な不活化の効果も確認し新たなPIPの標的候補と考えられた。
