Outline of Annual Research Achievements |
We analyzed the metabolic process of SN-38 nanoprodrug in cancer cells using the ZIF8-AgNW endoscopic probe developed in FY2022. The interaction between the released drug molecule SN-38 and intracellular substances was investigated by single cell SERS endoscopy. It was found that the SN-38 molecule was released from the nanoparticles within 3 to 6 hours after the particles were taken up by the cells, and penetrated into the cytoplasm and nucleus. However, even though no Raman signal from the SN-38 molecule alone was detected after 24 hours, cell death occurred after 24 to 48 hours, suggesting that SN-38 interacts with DNA and topoisomerase after 24 hours. The results reported here show that the developed endoscopy technique may play an important role in studying drug mechanisms of action.
