Outline of Annual Research Achievements |
The ability and working mechanism of visible and near-infrared (Vis-NIR) spectroscopy to estimate broccoli freshness were investigated. The Vis-NIR spectra of different broccoli parts were collected as predictors, and the cumulative temperature of broccoli under different storage conditions was determined as an indicator of freshness. By combining PLSR analysis and the stepwise selectivity ratio method, an accurate predictive model for broccoli florets freshness was constructed using informative wavelengths. Seven freshness marker metabolite candidates such as phenylalanine were identified by NMR-based metabolomics, which could explain the change in NIR absorption chosen for building the freshness prediction model.

Reason
As a JSPS international postdoctoral fellow in the year 2021, I performed NIR spectroscopy measurements on komatsuna. Based on this established approach, broccoli experiments were carried out smoothly during the year 2022. With the support from co-researchers, one of the research targets that investigating the ability and working mechanism of visible and near-infrared spectroscopy to estimate broccoli freshness was completed. Compared to the results of the previous komatsuna study, we made promising findings. The presentations at conferences received good responses from other researchers who are interested in the same topic. One paper is to be published. Therefore, the research is progressing smoothly according to the proposal.

Strategy for Future Research Activity |
Vis-NIRS and FF complement each other since Vis-NIRS has wider target molecules while FF has a higher sensitivity. After the development of estimating the freshness of komatsuna and broccoli using Vis-NIR spectroscopy, the study on estimating vegetable freshness using fluorescence fingerprint will be continued. Based on the further implementations of NMR-based metabolomics on the same vegetable samples, the common freshness marker metabolites and useful spectral wavelengths shared by different vegetables are expected to be clarified for building reliable freshness prediction models.

Causes of Carryover |
Research materials including vegetables and chemical reagents will be purchased for light-spectroscopic measurements and NMR analysis. Data analysis requires updating MATLAB and PLS_toolbox to use the latest formula functions. In the process of light-spectroscopic measurements, optical accessories such as lenses and optical fibers need to be replaced according to the characteristics of research materials and the experimental setup. Then, the conference fee and proofreading fee for publication are needed, as there is a need to exchange NIR research results and build relationships with other researchers through conferences and publications. Currently, there is a manuscript in need of proofreading, and an oral presentation is planned at the international NIR conference.
