Outline of Final Research Achievements |
In this study, we investigate the relationship between AT1R-associated protein (ATRAP) and glutaminase (GLS) and its pathophysiological significance in age-related renal injury at the cellular and biological levels. First, we focused on the relationship between ATRAP and glutamine metabolism. Using immunoprecipitation assays, we examined the molecular interaction between ATRAP and glutamine transporters at the cellular level, and analyzed the expression of glutamine transporters in ATRAP systemically overexpressing transgenic mice. These results are gradually clarifying the mechanism of anti-aging effects of ATRAP.

Academic Significance and Societal Importance of the Research Achievements |
未曾有の高齢社会を経験している本邦において健康寿命延伸の関心は高く,特に加齢性腎障害の克服は抗加齢対策の要となりえる.これまでに当研究室はATRAP欠損が加齢性腎障害を促進させることを示し,ATRAPの抗老化作用の可能性を明らかにした.近年,GLSは新規老化関連因子として報告されている.本研究では,ATRAPとGLSの関連性を明らかにし,ATRAPの抗老化作用の機構を探求することが目的である.これまでに細胞と生体レベルでATRAPとGLSの関連性を示す結果が少しずつ明らかになりつつある.しかし,十分にその関連性を証明するためには今後も継続した研究の必要性がある.
