Outline of Final Research Achievements

We investigated the relationship between environmental factors such as hypoxia and Hippo-Yap/Taz signaling pathway during tooth development. First, as a preliminary experiment, we determined the oxygen concentration and duration of the hypoxic condition. The results of organ culture of tooth germs under hypoxic conditions showed that the size of tooth germs was smaller and cusp formation was suppressed under hypoxic conditions. The results also suggest that, different from Hippo-Yap/Taz signaling pathway, superoxide dismutases (SODs) may also play important role in tooth development.

Academic Significance and Societal Importance of the Research Achievements

Hippo-Yap/Tazシグナル伝達経路は、細胞増殖を調節することによって、様々な組織や器官の大きさを制御することが知られている。本シグナル制御機構の異常はがんの発症や悪性化のみならず、歯胚の発生異常を惹起することが報告されているが、その詳細は明らかではない。本研究は、歯胚発生過程におけるHippo-Yap/Tazシグナル伝達経路関連遺伝子の機能解析および、母胎内の環境要因が歯胚発生に与える影響を解明することを目的としており、得られた知見は、歯胚の形態形成に関する基礎的な意義のみでなく、将来的には歯科再生医療の実現化に向けての一助となるなど、臨床的意義ももつと考えられる。
