Outline of Annual Research Achievements |
We are going on with the development of the lab-on-a-CD hardware in Takei laboratory in Nishi Chiba campus of Chiba University. Takei laboratory in Nishi Chiba campus of Chiba University. At the moment, we have managed to assemble the main components of the lab-on-a-CD hardware and improving the stability of the hardware to a robust form that can be easily transported between Takei laboratory in Nishi Chiba campus of Chiba University to the infection and host defense laboratory in Inohana campus of Chiba University where we shall be doing experiments using cultured malaria parasites. Since malaria is not endemic in Japan, the robustness will also enable the lab-on-a-CD hardware to be easily transported to Kenya where we are doing a collaborative research with Jomo Kenyatta University Of Agriculture and Technology (JKUAT) as well the institute of Primate Research (IPR). We are also doing in-vitro sensor analysis using Normal saline at increasing concentration to determine the functionality of each sensor. We are also using a mixture of chicken blood to represent malaria blood mixed with porcine blood to represent normal human blood to check if the hardware can help to determine the volume fraction of chicken blood in the porcine blood. These initial in-vitro studies will help to identify the major problems in the various components of the hardware such as in the 8- sensor array system or the Arduino or any other part that may cause the inconsistences in the impedance measurement results using the novel lab-on-a-CD hardware in diagnosis of malaria.

Reason
As mentioned in the earlier section, at the moment, we have managed to assemble the main components of the lab-on-a-CD hardware and we are going on with further modifications hence better the development of the lab-on-the-CD hardware for diagnosis of malaria. As at present, the lab-on-the-CD consists of an Arduino for controlling impedance measurement, an impedance converter, two high-speed analogue multiplexers and an 8-electrode sensor array for holding the samples thus enabling measuring 8 different samples at once. This helps to save on measurement time as well as enhancing repressibility of results. The sensors are connected on a compact disc which can be connected to a motor to enable centrifugation of the samples as needed to separate the different species of parasites in the samples.

Strategy for Future Research Activity |
Our plan for 2023 is to improve the lab-on-the-CD hardware based on a back-and-forth process of improving the hardware in Takei laboratory in Nishi Chiba campus and testing it on the cultured malaria parasites in the infection and host defense laboratory in Inohana campus. We plan to improve many aspects such as widening the frequency ranges of the hardware as well as the maximum rotation speeds during blood centrifugation by the lab-on-the-CD hardware. After confirming the working of the hardware, we plan to send it to Kenya for field trials using real situation of malaria infections. This will be done by the Kenya team. During this period, we will also do the data analysis of the experiments done using the developed lab-on-the-CD hardware, write a manuscript and submit to the journal for publication.
