Research Abstract

本年度の主な成果は以下のとおりである。
(1)マラリア感染赤血球の接着特性のシミュレーションについて、第一に、接着プロセスの第一ステップであるマラリア感染赤血球が血管壁近傍へと移動する現象のメカニズムを明らかにした。第二に、大規模並列計算のための、微小循環シミュレーション手法を構築した。第三に、原子間力顕微鏡を用いた接着因子の分子レベルでの接着・離脱特性を導入するため、分子間の接着・離脱反応を確立的に記述する接着モデルを導入した。
(2)血小板接着因子のシミュレーションについて、第一に、狭窄血管における血小板血栓形成のプロセスを明らかにした。第二に、血小板血栓と赤血球の相互作用について解析を実施した。
(3)細胞スケールにおけるシミュレーション技術の開発について、第一に、細胞膜モデルとしてのバネモデルと連続体モデルの振る舞いを解析的に比較検討した。第二に、せん断速度の大きさによって赤血球の配向が変化することを発見し、このメカニズムを振動伸張流れを用いて説明した。第三に、共焦点micro-PIVシステムを用いた観察により、微小血管分岐部と合流部における赤血球挙動の違いを明らかにした。
(4)組織・臓器スケールのシミュレーション技術の開発について、第一に、呼吸器系の複雑な管内流れをGPUを用いて大規模並列計算するための手法として、適合サブドメイン法を開発し、実形状肺内気流の計算を用いてこの有効性を実証した。第二に、共焦点micro-PIVシステムを用いた実験によって大腸菌の集団遊泳とエネルギ輸送を解明し、また、小腸内での微生物および栄養素の輸送について計算シミュレーションを用いてこれらを解析した。
(5)電子顕微鏡構造解析を用いて、気管上皮細胞の繊毛細胞の三次元構造を解明した。
(6)慣性を応用し、血液からがん細胞を分取するバイオチップなどを開発した。
これらの成果は、雑誌論文、および国際会議論文として報告している。

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

本年度は震災の影響で研究開始時期が大幅に遅れたにも関わらず、当初の計画以上の研究を実施することができた。今年度の研究計画は、主に分子スケールおよび細胞スケールのシミュレーション技術の開発と実験による検証であり、組織・臓器スケールのシミュレーション技術の開発は次年度以降に実施する予定であった。しかしながら既に、大規模並列GPU計算を用いた組織・臓器スケールのシミュレーション技術の基盤を構築することに成功した。また、同様に次年度以降に実施する予定であった細胞の生体力学を応用した診断用バイオチップについても、がん細胞を血液から分取する慣性を応用したチップの開発に成功している。これらが、当初の計画以上に進展していると評価する理由である。

Strategy for Future Research Activity

当初計画どおり、当面は分子スケールから臓器スケールまでの統合シミュレーション技術の開発を推進する。特に以下の項目について推進する。
(1)マラリア感染赤血球や血小板血栓に関わる接着分子のナノスケールの運動を導入した、微小循環シミュレーション技術の開発
(2)気管繊毛のナノスケールの構造・運動を導入した、気道上皮細胞がつくる流れのシミュレーション技術の開発。
(3)赤血球膜のナノスケールの分子運動を導入した、赤血球の新しい細胞モデルの開発と組織・臓器スケールシミュレーションとの統合。
