造血および白血病幹細胞ニッチの分子機構とその制御

Research Abstract

造血幹細胞ニッチをより詳細に解析するため、昨年に引き続き、炎症における造血幹細胞の動態を検討した。
cyclic-di-GMP (c-di-GMP)は、大腸菌やサルモネラ菌から産生される核酸で、この分子が造血幹細胞およびそのニッチにどのような影響を与えるかを検討した。
i)c-di-GMP(cdG)投与により、マウス骨髄造血幹細胞は減少しその機能も低下した。一方、脾臓では、幹細胞の増加が認められた。これにより、幹細胞の骨髄からの移動が考えられた。
ii) c-d-GMPの上記の効果は、cdGの細胞内受容体であるSTING欠失マウスでは見られなかった。iii) Angpt-1,Kitlのようなニッチ因子は、骨髄間葉系細胞で低下し、血管内皮細胞で増加していた。ゆえに、病原体由来のc-d-GMPは、幹細胞とニッチへの両作用により、造血を変動させることが明らかとなった。 これらの研究を通して「炎症性造血ニッチ」の実態が明らかとなった.また、これに類似して、CMLの造血においても、サイトカインによる造血ニッチの異常が起きていることを確認した[Kobayashi CI,et al: Blood, in press]。
