記憶想起後の記憶フェーズ決定機構の遺伝学的解明

Research Abstract

記憶想起後には、記憶を維持させる再固定化と、逆に記憶を減弱させる消去の二つの相反するプロセスが誘導される。これまでの解析から、再固定化及び消去誘導時には、前頭前野、扁桃体において遺伝子発現が誘導され、一方で、海馬では再固定化誘導時にのみ遺伝子発現が誘導されることを示し、これら3つの脳領域が再固定化及び消去において重要な役割を担うことを示唆してきた。本課題では、想起後に再固定化及び消去などの記憶フェーズの誘導が決定されるメカニズムの解明を目指す。本年度は、蛍光二重免疫染色法を用いてcAMP-responsive element-binding protein(CREB)及びextracellular signal-regulated kinase(Erk)の活性化マーカーとなるリン酸化を検出することで、海馬、扁桃体、前頭前野の脳領域において再固定化及び消去を制御し、生化学的に差異を示すニューロン集団の同定を進めた。その結果、再固定化フェーズでは海馬、扁桃体、前頭前野においてCREBのリン酸化のみが誘導された1種類のニューロン集団(pCREB+/pErk-)が出現することが明らかとなった。一方、消去フェーズでは、前頭前野では、CREBあるいはErkの一方のみのリン酸化が検出されるニューロン(pCREB+/pErk-あるいはpCREB-/pErk+)と、両者のリン酸化が共に検出されるニューロン(pCREB+/pErk+)の計3種類のニューロン集団、扁桃体では、pCREB+/pErk-ニューロンとpCREB+/pErk+ニューロンの計2種類のニューロン集団、海馬では、pCREB-/pErk+ニューロン集団のみの出現が観察された。今後、これらのニューロン集団の生化学的・機能的差異の解析を進める。また、海馬、扁桃体、前頭前野の脳領域の一つをリドカイン注入で不活性化することで三領域の相互作用を解析し、再固定化フェーズでは、扁桃体と前頭前野が海馬機能をコントロールすることが示唆された。
