癌転移におけるコンドロイチン硫酸の役割に関する分子メカニズムの解明

Research Abstract

以前我々は、コンドロイチン硫酸鎖中のE-unit構造(GlcA-GalNAc(4S,6S))を認識するファージ抗体が、マウスLewis肺癌細胞を強く染色し、その細胞の肺への転移がこの抗体によって、著しく阻害されることを見出していた。また、興味深いことに、種類の異なる多くのヒトの癌組織がこの抗体で強く染色された。そこで本研究では、これらの癌の転移の分子メカニズムにおけるコンドロイチン硫酸の役割を解明し、コンドロイチン硫酸を用いた創薬の可能性を追求した。その結果、ごく最近、肺癌細胞の転移に関わるコンドロイチン硫酸(CS)鎖の受容体として,Receptor for Advanced Glycation End products (RAGE)を同定した。さらに、抗RAGE抗体で、肺癌細胞の転移が有意に阻害されることも見出した。また、コンドロイチン硫酸鎖の合成に関わるN-アセチルガラクトサミン4-硫酸-6-0-硫酸基転移酵素の遺伝子発現をノックダウンすると、肺への癌細胞の転移が抑制されることも見出した。これらの知見は、コンドロイチン硫酸の癌転移における役割の解明だけでなく、将来的な抗癌剤の開発に繋がるものと考えられる。さらに、上述したE-unitを多く含むイカ軟骨由来のCS鎖およびエビ頭部由来のヘパラン硫酸が、日本脳炎ウイルスの感染を阻害できることも発見し、抗ウイルス薬の開発に向けて、一歩を踏み出した。
コンドロイチン硫酸鎖の生合成に関わるグルクロン酸転移酵素(GlcAT-I)の遺伝子変異が、ラーセン症候群を引き起こす遺伝病も同定した。また、コンドロイチン硫酸鎖中のグルクロン酸をイズロン酸に異性化する、デルマタン硫酸合成反応に関わり、双極性躁鬱病の原因遺伝子と推察されるデルマタン硫酸エピメラーゼ2(DSE2)の脳における遺伝子発現制御と糖鎖構造変化の相関を明らかにした。

Reason

当初の計画以上に進展している理由としては、査読有りの学術雑誌10編に論文が受理されたこと、および学会での発表も14件にのぼるからである。そのうち1報はCell Pressの姉妹誌の「American Journal of Human Genetics」に掲載された。また、世界的に有名な「Journal of Biological Chemistry」に2報掲載された。さらに、研究内容についても日本人の死因の1位を占める「がん」をターゲットとし、これまでほとんど解析されてこなかったコンドロイチン硫酸のがん転移における役割を明らかにしつつある。具体的には肺の転移に関わるコンドロイチン硫酸の受容体としてReceptor for Advanced Glycation End products (RAGE)を同定し、さらなる研究の発展が期待される。また、コンドロイチン硫酸あるいはその抗体、抗RAGE抗体に、肺への癌細胞の転移阻害活性も見出したことから、将来的な抗癌剤の開発に繋がることも期待できる。

Strategy for Future Research Activity

本年度の研究成果を踏まえ、今後さらに「癌におけるコンドロイチン硫酸の役割」について追究し、将来的な抗癌剤への開発に向けた研究を推進する。具体的には、(1)癌細胞を用いたコンドロイチン硫酸鎖生合成酵素遺伝子発現の網羅的解析:(2)siRNAによる癌細胞の転移阻害実験:(3)癌細胞を用いたコンドロイチン硫酸鎖の硫酸化構造の網羅的解析:(4)高転移性癌細胞表面のコンドロイチン硫酸-Eプロテオグリカンのコアタンパク質の同定:(5)高転移性癌細胞のコンドロイチン硫酸-Eプロテオグリカンの各種ヒト癌組織での発現スペクトルの解析などである。
