次世代肺がんチロシンキナーゼ阻害薬の感受性マーカーの同定と治療標的分子の探索

Research Abstract

われわれはALKのキナーゼドメインと相同性の高い30遺伝子を,EGFR突然変異およびK-Ras突然変異のない肺腺がん切除組織において定量的PCRによって発現解析を行った.そのなかで可溶型(非受容体型)チロシンキナーゼFERの過剰発現を3例で認めた.FERの過剰発現の理由としては,5'RACE法により隣接するPJA2遺伝子とのキメラトランスクリプトを認めたため,染色体転座の可能性についても検索を行ったが,過剰発現症例では染色体転座は認められず,トランプスプライシングによるキメラトランスクリプトであると結論した.スクリーニングで用いた症例を含む135例でFER免疫染色を行ったところ,原発性肺がんの約10%でFERの過剰発現を認めた.FER過剰発現症例は,明らかに無再発生存期間,全生存期間とも短く予後不良であった.多変量解析ではFER過剰発現とT因子,N因子は独立した予後不良因子であった.また,FERを過剰発現している肺がん細胞株を検索したところ,NCI-H661細胞でFERの過剰発現を認めた.ヘアピンsiRNAを用いて,FERの発現を抑制する系を構築し,FERの発現抑制の細胞への影響を検討した.NCI-H661細胞でsiRNAを用いてFERをノックダウンすると細胞がアポトーシスを来たし,治療標的候補となり得ることを見いだした.また,FER/PJA2のキメラトランスクリプトはFER過剰発現症例で認められ,普遍的に発現しているPJA2と過剰発現したFERとの間でキメラトランスクリプトが形成され,FERの過剰発現のマーカーとなり得ることを明らかにし,併せて発表した.
