腸内細菌依存性制御細胞による自己免疫制御に関する研究

Research Abstract

Mucosal-Associated Invariant T細胞(MAIT細胞)は、腸管に多く存在する自然リンパ球と考えられていたが、2009年のLantzらの報告により、ヒト末梢血においてもalpha betaT細胞の数％をしめる細胞群であることがわかり、免疫応答に重要であることが示唆された。MAIT細胞のヒトの自己免疫病態における役割は不明であるため、ヒトMAIT細胞の検出抗体の作製により、膠原病をはじめとする自己免疫疾患におけるMAIT細胞の動態や機能を解明し、病因解明や新規治療法開発の基盤を確立することを目的とした。
MAIT細胞T細胞受容体に対するモノクローナル抗体の作製については、バキュロウィルス発現システムを用いBombix moriによるTCR (Valha;7.2-Vbeta13)蛋白合成し、Balb/cマウスへMAIT-TCR組み換え蛋白（Valpha7.2-Vbeta13のヘテロダイマー）を免疫し、ハイブリドーマ作成を試みた。現在、数クローンの候補を得て現在解析中である。また、共同研究者からヒトMAIT細胞を検出できる抗体を得たので、それを用いてヒトMAIT細胞についての検討を行った。MAIT細胞は、健常人では末梢血alpha betaT細胞の数パーセントを占める細胞であり、主にCD8もしくはCD4,CD8double negative であった。全身性エリテマトーデス末梢血では、MAIT細胞の頻度が著しく減少していた。細胞増殖の低下とともに細胞死が増加しており、末梢血での低下の原因と考えられた。自己免疫疾患患者の腸内細菌叢は現在解析中である。
