Research Abstract

小胞体内で作られた各種成長因子、ホルモン、酵素類の外分泌や、受容体類は、全てゴルジ体へ運ばれ、それぞれの目的地へと輸送される。ゴルジ体は、小胞体から送られたタンパク質に、糖鎖修飾に代表される翻訳後修飾を施す。申請者らは、ゴルジ体のゴルジンタンパク質群の一つであるGiantinをノックダウンすることにより、分泌酵素の輸送が増大することを見出した。本研究では、1) この分泌増大のメカニズムを明らかにし、2) Giantin siRNAの、分泌機能改善薬としての実用化の可能性を追求する。現在、上記の目的を達成するためのデータを得るための解析方法を確立し、データを得はじめたところであり、おおむね計画通りである。
