| Research Abstract |
本研究においては、生理機能の不明なオーファン硫酸転移酵素(SULT)のSULT4A1およびSULT7A1に絞り込み、生理機能の解明を中心に研究を実施した。
平成25年度は、SULT4A1の機能解明に向けた新たな糸口を得るため、ゼブラフィッシュ胚を用いたホールマウントin situ ハイブリダイゼーション法による発生段階での遺伝子発現解析を行った。その結果、SULT4A1は脳全体および脊髄において発現していることを確認することが出来た。また、ノックダウン実験において、前脳、中脳、後脳の形態的異常が観察された。受精後6時間胚においては、既に神経系分化の予定運命が決まっており、今回RT-PCRでSULT4A1の発現を確認したところ受精後3時間胚では発現していないが、6時間胚では発現していることが確認出来た。このことから、ノックダウン実験における形態的発達の異常は、SULT4A1遺伝子のノックダウンが大きく影響して起こったものであるということが示唆され、SULT4A1が神経系の発生・発達においてなんらかの機能を担っていることが考えられた。
SULT7A1に関しては、平成25年度は、SULT7A1の基質特異性を詳しく調べたところ、PGE2やPGD2といったプロスタグランジンではなく、それらの代謝物であるシクロペンテノン型のプロスタグランジン(PGA2や15d-PGJ2)を硫酸化していることが明らかとなり、15d-PGJ2の硫酸体調製法の確立を行った。また、15d-PGJ2硫酸体の生理機能解明のため、免疫担当細胞である白血球T細胞やマクロファージの細胞株にプロスタグランジン硫酸体を作用させ、その影響を蛍光ディファレンシャル二次元電気泳動により解析した。その結果、15d-PGJ2硫酸体は、免疫担当細胞に作用し、その免疫機能に何らかの影響を与えていることが明らかとなった。
