AKB14-3-3-1(AKB1)の最適基質認識抗体の同定とinhibitor screeningの結果
AKB1のin vivoでの活性をよりよく反映するのは、去年報告したAKT substrate motif抗体よりもAMPK substrate抗体であることが判明した。V5タグを付加したTb14-3-3とAKB1を共にoverexpressionした細胞、およびV5タグを付加したTb14-3-3をoverexpressionし,かつAKB1をknockdownした細胞をV5-Tag抗体で免疫沈降した。Input, Bound(Immunoprecipitated), UnboundにわけてAMPK substrate motif抗体でWBすると、AKB1でリン酸化されたほとんどの蛋白はTb14-3-3に結合した状態で存在することが判明した。AKB1又はTb14-3-3をknockdownあるいはoverexpressionするとcytokinesisの異常がおこりmultinucleated 細胞が生じるが、このdetergent soluble なAKB1 Tb14-3-3 complexの量あるいは質的変化がそのような変化を起こしているのかもしれない。
Inhibitorに関してはPrem. Reddyの研究室でscreeningをおこなうも、nanomolar orderで効くAKB1 inhibitorは得ることができなかった。ところが100mMのHEPESおよびPIPES bufferがAKB1の活性を完全に抑制することが判明し、そのPiperazine ringが 活性を抑制することを見いだした。今後、Piperzaine ringをたくさん持つ化合物をscreeningする予定である。
