Research Abstract

natural occurring Th17(nTh17)細胞と腸炎惹起性のTh17細胞、In VitroでIL-6,TGF-bにより誘導されるTh17細胞、さらにIL-17Aを産生するILC17細胞の違いを明らかにしnTh17細胞についてはCD45RBhighのナイーブT細胞と共に移入を行うと腸炎を抑制する能力があることを発見した。驚くべきことにnTh17細胞はその細胞自身がTregとなっているのではなく、腸管にホーミングしたのちにNaive T細胞をTregに誘導することがわかり、現在現象のみしかとらえられていないが、今後そのメカニズムの解析が必要である。
本研究はTh17細胞のPhenotypeを詳細に分析したとして2012年Gastroenterologyに掲載され、その後2013年Annual review of pathology, Nature immunologyなどで引用される論文となっている。2011年のGastroenterology掲載後、命題となっている、腸炎にはどのタイプの細胞は必要であるかについては、現在Th17を経由するAlternative PathwayおよびTh17細胞を経由しないClassical Pathwayのどちらが腸炎に必要な因子であるのかについて詳細な解析を行っており、論文投稿を予定している。さらに小腸にホーミングするTh17細胞と大腸にホーミングを行うTh17細胞には違いがあるが、詳細な解析は行われず、今後の検討となる。

Reason

Th17細胞のPhenotypeを詳細に分析したとして上記の研究については、2011年に続き、2012年Gastroenterologyに掲載され、その後2013年th17細胞の特徴を評価した論文として、Annual review of pathology, Nature immunologyなどで引用される論文となっている。

Strategy for Future Research Activity

Th17細胞の腸炎への関与を今後も検討していく。現在Th17を経由するAlternative PathwayおよびTh17細胞を経由しないClassical Pathwayのどちらが腸炎に必要な因子であるのかについて詳細な解析を行っている。Th17細胞の存在しないRORgtKOマウスのNaive T細胞をTh1細胞ににIn Vitroで分化させ免疫不全マウスに移入をすると腸炎を発症する。どのようなメカニズムで発症するのかについて検討をおこない論文投稿を予定している。さらに小腸にホーミングするTh17細胞と大腸にホーミングを行うTh17細胞には違いがあるが、詳細な解析は行われず、今後の検討となる。

Expenditure Plans for the Next FY Research Funding

試薬等の消耗品に80万円、実験器具等の修理に間接費24円を使用予定。また、H24年度の未使用額の発生は効率的な物品調達を行った結果であり、翌年度の消耗品購入に充てる予定である。
