| Research Abstract |
肥満、高血圧、糖尿病、脂質異常症等の危険因子の集積によるメタボリックシンドロームが、心筋梗塞や脳卒中等の動脈硬化疾患と関連している。脂質シャペロンファミリーのひとつである脂肪酸結合タンパクaP2 (FABP4/A-FABP)は、脂肪細胞とマクロファージに存在し、代謝および炎症反応の両側面からメタボリックシンドロームの成因に深く関わることが示唆されている。我々は、糖尿病および動脈硬化に対してaP2が新規の薬物治療ターゲットになりうることを示した。最近、分泌シグナルペプチドを有さないもののaP2が脂肪細胞から分泌されることが示された。さらには脂肪細胞/マクロファージ以外に末梢毛細管や傷害血管の血管内皮に異所性に発現する可能性が示唆された。しかしながら、それらの生理学的役割や意義については不明なままである。本研究の目的は、分泌型aP2および異所性aP2が血管内皮機能や血圧とどのように関連しているかを検討することである。臨床血液サンプルを用いて血清aP2濃度を測定した。aP2濃度は正常血圧者に比べ無治療の本態性高血圧患者で有意に高値であった。aP2濃度は有意に年齢、BMI、血圧、中性脂肪値と正に、グルコースクランプで測定したインスリン感受性の指標 M値と負に相関した。重回帰分析では、年齢、性別、BMI、中性脂肪値で補正してもM値および血圧が独立したaP2濃度の説明変数であった。このことからaP2濃度の上昇は血圧上昇に関連することが示唆された。マウス血管内皮傷害モデルにおいて、免疫組織学的解析から再生された血管内皮細胞にaP2が発現することを確認した。現在各種薬剤による効果とaP2の発現量について確認中である。

Reason
臨床サンプルで血清aP2濃度を測定し、統計学的解析を行った。血管内皮傷害モデルを作成し、傷害血管内皮の再生部位にaP2の発現を確認し、各種薬剤による効果を検討中である。輸入した各種欠損マウスの繁殖が十分にできるようになり、これらを使ったin vivo実験を開始している。また、aP2阻害薬を使用したin vivo実験を開始している。

主に「その他」で計上していた直接経費の残りは主に物品費として次年度に使用する予定である。1. Wild-type, aP2-/-, mal1-/-, aP2-/-mal1-/-マウスの血圧経過を経時的に検討する。また、塩分負荷や各種高血圧マウスモデル(アンジオテンシン負荷、腎血管クリップ狭窄、5/6腎摘等)を作成し、血圧の変動を観察する。2. Wild-type, aP2-/-, mal1-/-, aP2-/-mal1-/-マウスから分離培養した内皮細胞におけるFABPアイソフォームの発現の変化や炎症マーカー、血管作動性物質、酸化ストレスマーカーおよびインスリンシグナルを検討する。
