ＨＬＡ－ＤＰ分子特異的免疫応答の解析：新しい免疫療法開発に向けて

Research Abstract

DPを含む不適合HLA分子に対するTリンパ球反応と抗体反応を、ヒト検体を用いて解析した。まずフローサイトメトリーを用いて、白血病/リンパ腫細胞株（Raji、Jurkat等）におけるDP陽性細胞の割合は84～99%、白血病患者から提供を受けた腫瘍細胞におけるDP陽性細胞の割合は20～99%であることを確認した。DPを含む数座のHLAが不適合の関係にある提供者2名の末梢血単核球を試験管内で共培養することで、不適合DPを認識するCTLクローンの樹立を試みた。計7組の共培養を行い、24のTリンパ球ラインを分離し、さらにクローン化して詳細な解析を行った。しかし不適合class I分子や不適合DR分子を認識するCTLクローンは分離されたが、不適合DPを認識するクローンは分離されなかった。また血小板輸血不応性の血液疾患患者8名から血清の提供を受け、その中に存在する抗HLA抗体が認識するHLA分子の同定を試みたところ、不適合class I分子に対する抗体は検出されたが、不適合DP分子に対する抗体は検出できなかった。ちなみにDP以外のHLAを認識する抗体を含む患者血清を用いて解析したところ、抗HLA抗体の細胞傷害活性には少なくともADCC活性が関与することが確認された。以上より、HLA不適合造血幹細胞移植患者において、DP分子特異的免疫応答は他のHLAに対する免疫応答に比べて弱いものであろうと考察される。現在さらに各DPアリルのcDNA constructを作成しCOS細胞や.221細胞など他のHLA分子を発現してない細胞へ遺伝子導入し、その細胞を用いてTリンパ球を刺激する実験を進めている。
