呼吸器悪性腫瘍における自家がん特異的免疫応答を利用した新規腫瘍抗原の同定

Research Abstract

本研究では、呼吸器悪性腫瘍患者の自家がん特異的免疫応答を利用した新規腫瘍抗原の同定・解析を行うことにより、新たな診断、治療の標的となる分子の探索を行う。特に自家がん特異的免疫応答を利用するため、実際に癌患者生体内で生じている生体防御機構の解明にもつながる。平成23年度は、研究計画1)-3)に対して以下の結果を得た。
1)新たな肺大細胞癌の自己リンパ球-自己腫瘍細胞システムを用いて、リンパ球-腫瘍細胞混合培養を繰り返し、自家がん特異的細胞障害性T細胞のlineを誘導した。このbulk CTLから限界希釈法でCTL cloneを得られた5クローンの内、Clone 2-4を用いて、抗腫瘍活性を検討したところ、自家肺癌細胞に特異的に細胞障害性を示し、また他家の肺腺癌、肺扁平上皮癌を認識し、これらの活性は抗HLA-A24抗体で阻害された。
2) Clone 1を用いてcDNA発現クローニングの結果、陽性反応を示すcDNA cloneのアミノ酸配列から、HLA-A2402拘束性binding motifを有する5種類のアミノ酸を合成し、CTLとの反応性を検討した。Peptide1に反応性の上昇を認めたが、反応が弱いため、別の候補ペプチドを合成している。
3) 肺腺癌からわれわれが同定した癌精巣抗原である、KK-LC1の新規エピトープの探索のために、日本人に頻度の高いHLA-A2, A24にbinding scoreの高いHLA-A2ペプチドとHLA-A24ペプチドを、同患者の自己の末梢血単核球から誘導した樹状細胞にpulseし、凍結保存しておいたリンパ節リンパ球をresponderとして、共培養を繰り返したところHLA-A2拘束性のbulk CTLが誘導された。
