Research Abstract |
Amongst prostate cancer cell lines, we found Dkk1 was expressed in the culture supernatant of castrate-resistant PC-3 cells by western blot. We established LNCaP cell line inducibly expressing Dkk1 in a tetracycline dependent manner. However, those cells did not exhibit castration resistance in vitro by inducible Dkk1 expression. On the other hand, invasion of PC-3 in boyden chamber was significantly inhibited by Dkk1 knockdown by siRNA. These results suggest that Dkk1 expression does not directly contribute to acquisition of castration resistance but is rather involved in invasiveness of castrate resistant prostate cancer cells.
