滲出型、萎縮型加齢黄斑変性への小胞体ストレスの関与とそれを標的とした治療法の開発

Research Abstract

加齢黄斑変性眼の入手先はサンディエゴアイバンクに変更し、加齢黄斑変性眼を入手し次第の連絡を依頼していたものの、先方のテクニシャンの退職などに伴い、業務が停止していた。26年になって、ようやく先方の体制も整い、3月後半になって加齢黄斑変性眼が入手できたとの連絡があり、パラフォルムアルデヒド固定の後に郵送するよう依頼した。
またジャクソンラボラトリーより、カベオリン１ノックアウトマウスのつがいを購入した。購入当初は母親が子供を育てないため死んでしまうという問題があったが、これは母の脳へのアミロイドβの蓄積によるのではないかと考え、若い母親に育てさせたところ、問題なく発育することが確認できた。その後、ノックアウトマウスの数を増やしているところである。
また、視細胞特異的カベオリン１ノックアウトマウスの眼球を入手し、電子顕微鏡を用いてその観察を行った。視細胞外節、神経終末のリボンシナプスを含めて大きな異常は確認できなかった。
