ヒト皮膚由来の多能性幹細胞を用いた骨髄移植法の開発

Research Abstract

皮膚や骨髄などの成人ヒト間葉系組織あるいは培養間葉系細胞に多能性を有する新たな幹細胞 Muse細胞を見出した。この細胞はNanog, Oct3/4, Sox2などの多能性幹細胞マーカーを発現し、1細胞から3胚葉性の細胞に分化する能力を有し、自己複製能を持つ。ヒトES細胞のマーカーSSEA-3を発現し、このマーカーを用いることによりヒト線維芽細胞, 骨髄間葉系細胞の培養細胞の他に、新鮮骨髄液、皮膚などの生体組織からも直接分離可能である。また通常の成人ヒトの組織に存在し、腫瘍性がない。
本研究では多能性を有するが腫瘍性の無いMuse細胞の特性を骨髄移植に活用することを目的とし、皮膚(真皮)由来のMuse細胞から造血系細胞への分化の可能性を模索した。
本年度はX線照射を行った免疫不全マウスSCIDマウスへの移植を行った。lentivirus-GFPでラベルされたヒト皮膚線維芽細胞由来Muse細胞をマウスの尾静脈から投与した。投与した細胞数は1万細胞とした。6ヶ月後にGCSFを投与して投与されたマウスの末梢血を回収し、FACSで解析した。その結果、ごく少数のヒトGFP+細胞がGFP陽性細胞におけるpacific blue-CD45(白血球系マーカー)、PE-Cy7-CD2(T細胞リンパ球系マーカー)、APC-CD19(B細胞系)を発現していることが確認された。また骨髄内を組織学的に検討した結果、GFP陽性のMuse細胞が造血系細胞マーカー陽性の細胞として分化して骨髄内に生着していることが確認された。真皮は中胚葉由来組織ではあるものの、同じ中胚葉とはいえ造血系細胞は性質がかなり異なる。多能性のMuse細胞を使うことによって、造血細胞への分化の可能性が示唆されたと言える。
