肺および肝臓内の炎症細胞インビボライブイメージングシステムの構築

Research Abstract

本研究では申請者らが開発したマウス肺内in vivo live imagingシステムを用いて、喘息肺での種々のTh細胞分画（Th1/Th2/Th17/iTreg等）の肺内ダイナミクス解析を行った。またこれらの技術を応用して肝臓内in vivo live imagingシステムの構築を行い、以下の結果を得た。
1)マウス肺内in vivo live imagingシステムによる喘息肺でのリンパ球の動的挙動解析：抗原特異的な種々のTh細胞分画について喘息肺での浸潤様式の差異を解析したところ、Th1、Th17細胞などは肺組織内でTh2細胞のような明瞭な細胞集団を形成しにくいことが明らかとなった。さらにTh1やTh2細胞について抗原吸入後の肺組織内での動的挙動解析を行って差異を検討した。またステロイド剤を投与するとTh2細胞の肺への流入数が減少した。
2)ナイーブT細胞とエフェクターT細胞の浸潤様式の比較：ナイーブT細胞とエフェクターT細胞の浸潤様式を比較したところ、抗原吸入後エフェクターT細胞のみが肺に集積し、抗原特異的CD4T細胞であってもナイーブT細胞の場合には肺に集積しないことが明らかとなった。
3)遺伝子操作マウスのT細胞を用いた実験：マウス喘息モデルで気道炎症が起きないことが示唆されているCD69ノックアウトマウスのTh2細胞について肺への集積能を検討したところ、有意に集積能が低下していた。
4) 肝臓内in vivo live imagingシステムの樹立：我々の樹立した肺内in vivo live imagingシステムの撮影技術を応用して、これまで敬遠されてきた肝臓内のライブイメージング実験系を確立した。ConA肝炎モデルを用いてリンパ球の浸潤様式を解析したところ、ConA投与12時間でCD4T細胞や好中球が肝臓内に激しく集積して炎症が誘導されることが明らかとなった。
