| Research Abstract |
アレルギーになりやすい、なりにくいと言う体質は、遺伝的にプログラムされている。本研究ではIL-4遺伝子の3’側非転写領域に存在する3カ所のSNPsに着目し、SNPをゲノムに遺伝子導入したマウスを作成することで、その生物学的意義を解明していく。IL-4遺伝子の3’側非転写領域 HS4にアトピー患者で多く見られるSNPがあることを見出した。そこで、非アトピータイプSNP ヒトサイレンサーノックインマウスとアトピータイプSNPヒトサイレンサーノックインマウスを確立した。ヒトHS4ノックインマウスについて卵白アルブミン（OVA）特異的免疫反応を解析するため、それぞれのノックインマウスでOVA特異的T細胞抗原受容体（TCR）を持つトランスジェニックマウスを作成してTh2への分化とIL-4産生を測定した。マウスでサイレンサーとして同定されたHS4領域は、ヒトにおいても配列は良く保存されていいた。しかしながら、機能としてはサイレンサー活性を持たないことが、ノックインマウスの解析から明らかにされた。また、アトピータイプSNPを持つノックインマウスは非アトピータイプに比べ､僅かにではあるが高いIL-4産生能を有していることが証明された。この結果より、アトピー患者で認められたサイレンサー領域に存在するSNPは、T細胞におけるIL-4産生を亢進する働きがあることが､マウスの生体を使った機能解析において証明された。
