Outline of Annual Research Achievements

AChE阻害作用に基づく薬剤は,脳内AChEのバランスを整えることで記憶障害の改善を図る対象治療薬となるが,その誘導体に神経成長因子促進作用の増強を見い出すことができれば,アルツハイマー病等の神経変異疾患に対する根治治療薬の開発に繋がることが大いに期待される。本研究では,その根治治療を目指した化合物シードの創出を指向し,本年度は,主として,ガランタミンの生合成に関わる酵素の中で,4-O-メチル化酵素(4OMT)の触媒機構の解明を目指し,4位と3位へのメチル化効率の異なる3種の4OMT,Narcissus pseudonarcissus由来norblladine 4OMT(NpN4OMT),Lycoris longituba由来OMT(LlOMT),及びLycoris aurea由来OMT(LaOMT)について結晶化を進めた。その結果,いずれにおいても,3,4-ジヒドロキシベンズアルデヒド(1)とSAMを複合したX線結晶構造の取得に成功した。これにより,LlOMTとLaOMTは,1の4位水酸基を触媒残基Lys158と相互作用できる距離に結合できるのみならず,活性中心キャビティー内でその結合が反転した形でも結合することができるため,1の4位と3位水酸基を各々メチル化できることが判明した。さらにこれら4OMTの活性中心キャビティ-を比較することで,1の4位水酸基のみをメチル化するNpN4¢OMTにおいては,52番目と186番目のアミノ酸(Ser52/Tyr186)がLIOMT(Met52/Phe186)とLaOMT(Thr52/Phe186)とは明らかに異なっていることを見いだした。そこで次に,これらについて変異導入を行った結果,4OMTは,これらのアミノ酸の違いにより,1の4位と3位水酸基へのメチル化効率を制御していることが明らかになった。

Reason

当初の計画通りに、4OMTの結晶構造の取得と基質水酸基へのメチル化位置選択性の制御について明らかにすることができたことから、総合的に概ね計画通りに進展していると考えている。

Strategy for Future Research Activity

本年度において得られた4OMTの触媒機構に関する知見を踏まえ,4OMTの機能改変およびそれら機能改変酵素を用いた新規化合物群の創出を目指す。また,フペルジンAの生合成や4OMT以外のガランタミン生合成に関わる生合成酵素についても触媒機構の解明と機能改変の取り組むことで,さらなる新規化合物群の創出を試みる。
