Outline of Annual Research Achievements

肥満に随伴する酸化ストレス障害は,骨格筋の劣化を介した筋-肝連関により非アルコール性脂肪性肝炎 (NASH) の発症と進展を促進し,肝発癌のリスクを増大させる.本研究では,生体の抗酸化ストレス応答の司令塔因子Nrf2に着目し,NASH-肝癌の抑止における筋Nrf2の役割を解明する.本年度は以下の実験を実施した.
筋特異的Nrf2遺伝子レスキューマウスの作製と表現型解析: 全身Nrf2遺伝子欠失マウス (Nrf2-KO) をベースに,筋にのみNrf2を発現する筋特異的Nrf2遺伝子レスキューマウス (Nrf2-mRes) を作製した.NASH誘導食である高脂肪果糖食の摂餌下で,Nrf2-KOとNrf2-mResの肝病態を比較した.Nrf2-KOでは,高度の肝線維化を伴う重症NASHを発症した.一方,Nrf2-mResではNASHの発症が強力に抑止されていた.肝組織におけるRNA-seq解析において,Nrf2-mResではNrf2-KOと比較して,炎症性サイトカインやケモカインの遺伝子発現が低下していた.Nrf2の制御下で骨格筋から肝臓へと抗炎症作用を有する物質が輸送されていることが示唆された.骨格筋では,筋へのNrf2遺伝子レスキューにより,遅筋 (Type 1) 線維の増加,ミトコンドリア含有量および活性の増大が確認されている.現在,上記マウスの骨格筋検体や培養筋細胞を用いて,Nrf2に制御され,NASHを抑止する筋由来の因子を同定すべく各種解析を継続している.

NASH-肝癌モデルp62:Nrf2 double knockをベースとした,筋特異的Nrf2遺伝子レスキューマウスの作製が完了した.

Strategy for Future Research Activity

現在,NASH-肝癌を誘導する食事介入を実施しており,次年度以降はNASH-肝癌の抑止における筋Nrf2の役割に関して,検証していく予定である.
