Outline of Annual Research Achievements

ラットに、手掛かり刺激の位置を短時間記憶に保持し、その後に同じ方向に反応させる遅延順反応課題と、逆の方向に反応させる遅延逆反応課題を、８試行ごとに切り替えて行うよう、訓練した。この課題を遂行中に、前頭葉および頭頂葉の広い領域からマルチニューロン活動を記録した。記録を行った領域の中で、とくに内側前頭皮質の背側部（dmPFC）において、遅延期間に持続的な活動を示すニューロンが多く分布していることが明らかになった。これらのニューロンに、独自に開発した電気穿孔プラスミド注入法によって蛍光タンパク質の遺伝子を導入して標識し、組織学的解析を行った。その結果、遅延期間に持続的活動を示すニューロンのほとんどは、３層あるいは５層の錐体細胞であり、それらの多くが線条体に投射していることが明らかになった。これらの結果より、遅延期間中の持続的発火活動には、皮質－基底核ループ回路が重要な役割を果たしていることが示唆された。

当初計画した実験のうち、ニューロン活動記録および課題関連ニューロンの標識および組織学的解析については、当初の予想を上回る結果が得られている。一方で、上記の実験に注力したため、イオントフォレシスの実験についてはまだ十分なデータが得られていない。

Strategy for Future Research Activity

ニューロン活動記録および課題関連ニューロンの標識および組織学的解析について、データ分析および論文執筆の作業を進める。イオントフォレシスについては、データ収集を加速させる。
