Outline of Annual Research Achievements

これまでに植物免疫を阻害する化合物を３個単離して、植物免疫ホルモンであるサリチル酸と構造的に類似していることを見いだしている。このうち免疫阻害剤P7が特異的に結合するタンパク質（CUPIN1と呼ぶ）を精製単離して同定している。抗体を用いた解析により、CUPIN１はアポプラストに存在することがわかった。また、その共結晶構造解析とアミノ酸残基の変異体解析により、P7と結合するポケットが存在することが明らかになった。サリチル酸添加時に観察されるNPR1の核内での蓄積が、P7によって阻害されること、さらにサリチル酸添加時に観察される細胞内の酸化還元(レドックス)状態の変化がP7によって阻害されることを示された。CRISPER-CASシステムを立ち上げ、CUPIN1遺伝子のホモログをKOしたところ、他のCUPIN1ホモログの発現上昇が確認されたことから、CUPIN1遺伝子群の補償作用が示唆された。また、複数の新規病原体デノボゲノム解析を行い、比較解析によりエフェクター候補遺伝子を同定した。
