加圧トレーニングによる骨格筋肥大の分子機構の解明と高齢者サルコペニアへの応用

Outline of Annual Research Achievements

前年度に引き続き、低強度電気刺激によるトレーニングのラット骨格筋各種蛋白質発現に及ぼす血流制限(加圧)の影響について解析した。[方法] ラットをコントロール群、電気刺激のみ群、加圧下電気刺激群、加圧のみの4群に分けて検討した。低強度電気刺激によるトレーニング (前脛骨筋にアイソメトリック収縮)を、週3回3週間実施し、トレーニング後に前脛骨筋を摘出して蛋白発現をWestern blotting法により検討した。血流制限は大腿部のカフ装着によって血流制限を加えた。[結果] コントロール群および加圧のみ群、電気刺激のみ群に比し、加圧下電気刺激では、GLUT4、PGC1αの有意な発現の亢進を認めた。コントロール群に比し、電気刺激のみ群、加圧下電気刺激群では、FNDC5、COX4, MCT1の有意な発現の亢進を認めた。MCT1の発現は、加圧群でより顕著であった。GLUT4, MCT1, FNDC5, COX4の発現とPGC1αの発現は有意な相関が見られた。[結語] 血流制限下(加圧下)での慢性低強度電気刺激によるトレーニングでは、GLUT4, PGC1α, COX4, MCT1などの発現が有意に亢進し、ミトコンドリア機能、糖代謝改善につながる可能性があり、今後、高強度のトレーニングが実施できない高齢者、廃用症候群などへの新たなるトレーニング法となる可能性があると考えられた。
