分類学を発展させるＤＮＡ情報を伴ったエピタイプ化推進－きのこ類・地衣類

Outline of Annual Research Achievements

博物館などに保管されているタイプ標本は生物の学名の基礎となっているものであり極めて重要な価値を持つ。しかし，近年ではDNA情報が分類学的研究に重要な要素となっており，古いタイプ標本からのDNA情報の収集が困難であることが研究の足かせになっている。本研究ではきのこ類および地衣類のタイプ標本採集地から該当分類群を採集し，DNA情報を伴ったエピタイプ標本を指定することによって，近代分類学における問題解決および博物館等に保管されているタイプ標本のさらなる価値向上を目指すものである。
平成26年度は次の研究を実施した。
①国立科学博物館所蔵日本産きのこ類・地衣類タイプ標本に該当する現行種のDNA解析。国立科学博物館に所蔵されている日本産きのこ類・地衣類タイプ標本について標本データを整理しデータベースを作成した。地衣類のタイプ標本は490分類群であるが，現在分類群は383分類群にまで整理されている。きのこ類タイプ標本は562分類群(680標本)を確認し，うち312標本からはDNAを抽出済みであるが，それらの大半は断片化が高度に進行しており，通常の方法で塩基配列が特定できたのは60標本のみであった。それら現行分類群のうち，タイプロカリティーから採集された当該標本からDNAが得られたものは，きのこ類30分類群，地衣類14分類群であった。DNAが得られた標本については，ITS rDNAやmtSSU, nrLSU, RPB1など菌類で主要な遺伝子領域を順次解析した。
②国際学会でのエピタイプ重要性についての発信。エピタイプ指定の重要性について，招待講演により第10回国際菌学会（タイ・バンコク開催）で地衣類に関する発表を行った．また，タイプ標本以来採集がなかった小笠原諸島産シンジュタケの再発見と新知見について，2件の招待講演（日本菌学会主催・日韓合同シンポジウム，第10回国際菌学会）にて発表した。

Reason

国立科学博物館では，日本から記載されたきのこ類のタイプ標本が562分類群（680標本），地衣類では490分類群（523点）を保管している．多くのタイプ標本採集地が採集当時の環境と比べて著しく破壊が進んでいることや記載分類群に分類学的に難解なものが含まれていることを考慮すると本プロジェクトの期間内にそれぞれ100分類群程度エピタイプ指定ができれば，プロジェクトは成功であると考えており，計画的にエピタイプ候補種の採集も進んでいる．一方，エピタイプ指定時に示すべきホロタイプまたはレクトタイプ中のDNA分断化状況の調査についても計画的に進んでいる．
また，我々の取り組みについては国際的にも注目されており，2014年度には地衣類・きのこ類それぞれで国際学会からの招待を受けて講演を行った．
以上より，本プロジェクトの進行状況および国際的評価の点でおおむね順調に進展していると考える．

Strategy for Future Research Activity

日本国内のタイプ標本採集地をまわりタイプ標本同等品の収集に努め，可能な標本からエピタイプ指定を行っていく．タイプ標本または同年代の採集品のDNA状態については国立科学博物館保管標本だけでなく，海外の標本庫の標本についても比較できるよう研究機関間のネットワーク構築に努める．

Causes of Carryover

プロジェクトを進めていく上で難解な分類群を効率よく検討するために海外研究者の招へいが必要だと感じ，その費用の確保を考えていた．当該年度は購入していた試薬が他予算で購入できたため，その分が次年度使用額として生じた．

Expenditure Plan for Carryover Budget

本研究担当者による日本各地での標本採集に加えて，海外研究者の招へいも積極的に行い，難解な分類群もより正確に同定できるような体制を取って研究を進める．従って，予算使用は，調査・採集，標本の形態・化学・遺伝子解析，そして海外研究者招へいおよび国際ネットワーク強化を基本とする．
