| Outline of Annual Research Achievements |
博物館等に保管されているタイプ標本は生物の学名の基礎となっているものであり極めて重要な価値を持つ。しかし，近年ではDNA情報が分類学的研究に重要な要素となっており，古いタイプ標本からのDNA情報の収集が困難であることが研究の足かせになっている。本研究ではきのこ類及び地衣類のタイプ標本採集地から該当分類群を採集し，DNA情報を伴ったエピタイプ標本を指定することによって，近代分類学における問題解決及び博物館等に保管されているタイプ標本のさらなる価値向上を目指すものである。
平成28年度は，北海道、宮城県仙台市周辺、茨城県、東京都（小笠原諸島）、京都・滋賀県、鹿児島（奄美大島）、沖縄（石垣島）等を調査し，きのこ類約1500点，地衣類110点の標本を得た。そのうち，きのこ類については，小笠原諸島から61標本、京都・滋賀から45標本、奄美大島から1標本、石垣島から2標本のエピタイプ候補標本をタイプロカリティーから得ることができた。同様に，地衣類は7種10標本のエピタイプ候補標本を得ることができた。本プロジェクトにおいてはこれまでに延べ現行種(245分類群)の標本を得た。DNAが得られた標本については，ITS rDNA, mtSSU, nrLSU, RPB1, RPB2, tef1, MCM7, ATP6といった菌類で主要な遺伝子領域を解析し、Genbankを通して公開予定である。一方、国立科学博物館および国内外その他標本庫所蔵の古いタイプ標本からのDNA情報の収集も試み、きのこ類では最長約75年前まで、地衣類では約15年前までの標本について、DNA部分配列の解読ができた。本プロジェクトでは、国立科学博物館所蔵タイプ標本のデータ整備もあわせて進め、ホームページ上にきのこ類100点のタイプ標本画像をすでに公開しており、地衣類についてもタイプ標本リストおよび100点の画像を公開予定である。
