絶滅危惧両生類における遺伝的多様性評価と保全のための包括的研究

Research Abstract

① マイクロサテライト(SSR)の開発。絶滅危惧両生類で南西諸島産のハナサキガエルおよびバングラデシュ産トラフガエルについて、SSRマーカーを開発した。
② 野外集団からのサンプリング。上記の絶滅危惧両生類について、沖縄島およびバングラデシュの多数の集団から網羅的なサンプリングを行なった。
③ 種ごとに適切な人工受精法と飼育法の確立。上記の各種について、人工受精と飼育条件の最適化を行なった。アマミハナサキガエルとオオハナサキガエルとトラフガエルについて、飼育下繁殖に成功した。
④ 精子凍結保存法の確立。南西諸島の絶滅危惧種のうちアマミアカガエルとアマミイシカワガエルとアマミハナサキガエルを用いて凍結保存液の至適化を行った。1~7日凍結保存後の精子を解凍し、精子の運動能を観察するとともに、人工受精法によって媒精を行い、受精率を測定したところ、7日凍結精子でも受精可能であることを確認した。
⑤ 集団構造の解明および集団内の遺伝的多様性と絶滅リスク評価。野外で採集した上記サンプルについてSSRマーカーを用いてジェノタイピングを行い、集団内の遺伝的多様性と、集団間の遺伝的構造を解明した。

いずれの項目についても実験を行っており、ほぼ計画通りに進展している。

Strategy for Future Research Activity

今年度も、ほぼ申請書に記載の計画で研究を推進していく予定である。特に以下の点については、昨年度の成果を検討しながら、重点的に実験を進めていく計画である。
マイクロサテライト(SSR)マーカの開発:南西諸島産絶滅危惧両生類(アマミハナサキガエル・オオハナサキガエル・コガタハナサキガエル)についても、SSRマーカを開発し、野外で採集したサンプルについてジェノタイピングを行い、各絶滅危惧両生類について、集団内の遺伝的多様性と、集団間の遺伝的構造を解明し、絶滅リスクを評価する。種ごとに適切な人工受精法と飼育法の確立:各種各集団について、人工受精と飼育条件の最適化を目指して、各種に適切な飼育法を確立する。精子凍結保存法:アマミイシカワガエルとアマミハナサキガエルを用いて凍結保存液の至適化を行い、各保存試薬の効率を評価するとともに、3ヶ月・半年・1年以降(H27年まで)に人工受精を行い、凍結保存可能期間を明らかにする。遺伝マーカによる血縁度測定と種親選定による計画的交配:飼育下繁殖個体を用いて、血縁度の異なるいろいろな組合せで人工交配を行い、それぞれの子孫について生活力を検証し、飼育下で遺伝的多様性を維持できる継代飼育システムを確立する。
