Outline of Annual Research Achievements

これまでに得られている概要ゲノムデータおよびトランスクリプトームデータから、－株ゲノムのみに存在し、転写因子をコードするCpMinus1遺伝子が得られている。CpMinus1のcDNAをHSP70遺伝子プロモーターの下流で強制発現するようなコンストラクトを作製し、＋株に導入し6株の形質転換体を得たところ、2株は＋株と混合することで顕著に接合反応が進行し、－型との混合ではほとんど接合子を形成せず、あたかも－型細胞のような表現型を示したものの、残りの株では完全に性表現の転換は起こらなかった。表現型が揃わない理由として、遺伝子発現に用いたプロモーターが不適切であることが考えられた。そこで本年度は、－型細胞のゲノム配列からCpMinus1遺伝子のプロモーターを含む領域を増幅し、新たな遺伝子導入用コンストラクトを作製し、再び＋型細胞へ遺伝子導入した。その結果、野生型－型細胞におけるCpMinus1遺伝子の発現に比べて、形質転換体では約10倍の発現レベルを示すようになった。これらは顕著に－型化傾向示したものの、依然として自家生殖能力を示した。またCpMinus1遺伝子の発現を抑制するために、antisense方向に挿入したコンストラクトを－型細胞へ遺伝子導入し、これらの形質転換体の表現型解析を試みた。antisense RNAでは、完全な機能抑制は期待できないためか、これらについては、一部の株で弱い自家生殖能力を示したが、－型細胞としての表現型に大きな変化は見られなかった。以上から、CpMinus1遺伝子の発現が十分であれば、顕著に－型細胞化するものの、①＋型細胞ゲノムに特異的に存在するCpPlus1遺伝子の機能と干渉する、または②－型細胞ゲノムに特異的に存在し、CpMinus1と協調的に働くCpMinus2遺伝子がないため、100％完全には性が切り替わらない可能性が示唆された。
