| Outline of Annual Research Achievements |
全エクソン配列解析の結果、平均カバレージ69.5, 10×カバレージ 平均93.8%のデータを得た。非同義性変異、挿入欠失を抽出し、1000 Genome, DB exomeデータベースで頻度1%未満または登録なし、バリエーション/カバレージ>0.3、クオリティバリュー>15を基準にフィルタリングし、一人当たり平均で一塩基置換1000個、挿入欠失315個を得た。Polyphen, SIFT, GERP等の機能予測解析、罹患者-対照者の比較検討を加え、複数の候補遺伝子を得た。例えばGene Aは複数の家系で罹患者に有意に集積し、一部ではノンセンス変異を来していた。機能解析のためRNA干渉、clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9法を用いて膵がん細胞におけるノックダウン、ノックアウトを行った。一部遺伝子についてノックダウン、ノックアウトにより細胞増殖が亢進することが観察された。東京女子医科大学病院を受診した膵がん患者で家族性膵がん該当例と非該当例を比較検討し、家族性膵がん該当例に50歳以下の若年者が多いことを見出した。集積した家族性膵がん家系では若年発症膵管内粘液性乳頭状腫瘍(IPMN)が多く認められた。IPMN関連遺伝子の解析、孤発例を加えた臨床病理学的徴候との関連について明らかにした。
