Outline of Annual Research Achievements

本研究は、従来ゲノムの観点のみから解析が行われていた低酸素応答について、エピゲノムの観点を取り入れて詳細な解析を行い、これを腎臓病学の病態生理に応用するものである。まず、培養血管内皮細胞を低酸素で刺激し、ヒストン修飾蛋白の発現変動を解析し、その変動に相当するヒストン修飾の変化を ChIP-seq で解析し、種々の分子の hypoxia inducible factor (HIF) 結合領域周辺の変化に注目して解析を進めた。これまでの研究では、嫌気的解糖を助ける glucose transporter 3 (SLC2A3) の低酸素状態におけるエピゲノミックな調節作用、低酸素状態の腎臓で最もシグナル・ノイズ比よく発現が増加する angiopoietin like 4 (ANGPTL4) のエピゲノミックな発現調節とそれにともなうクロマチン高次構造の変化などを解明してきた。また、その過程で施行した HIF の ChIP-seq で同定した新しい HIF target 分子である SPAG4 が細胞質分裂に関与し、その発現が腎臓癌の予後不良と相関することを示した。慢性腎臓病では低酸素が末期腎不全への final common pathway となっている。本来そのような腎臓では、HIF が低酸素に対する防御機構として発動するべきであるが、尿毒素によって HIF と co-factor である p300 との結合が阻害され、尿毒症状態ではヒストンのアセチル化が減弱することにより十分な生体防御作用を発揮できないことを示した。更に、HIF の新規調節分子を同定するために siRNA library を用いたスクリーニングを行い、炎症に関与する CEBPdelta が HIF の発現を mRNA level で調節していることを示し、低酸素と炎症の新しい相互関係の分子機序を示した。
