Outline of Annual Research Achievements

1)活性酸素産生酵素NOX1のより制御されるうつ様行動発現機構の解析
本年度は、機序解明としてNOX1由来ROSの標的分子の同定をレドックスプロテオミクスにより試みた。慢性コルチコステロン投与によりROS産生増加が認められた prefrontal cortex(PFC)の野生型マウス(WT)でNR1, NR2B, N-ethylmaleimide-sensitive factor(NSF)のシステイン残基の酸化修飾が認めれたが、NOX1遺伝子欠損マウス(NOX1-KO)においては、その修飾が抑制されていた。
2)恐怖記憶におけるNOX1の役割解析
fear-conditioning test にて解析を行った。恐怖記憶の獲得については WT と NOX1-KO で違いは認められなかった。しかし、恐怖記憶の消去学習について解析したところ、NOX1-KOで有意に増強されることが認められた。
