過酸化水素バイオイメージングのための高速・高感度過酸化水素蛍光プローブの開発

Outline of Annual Research Achievements

過酸化水素は生体に害を与える活性酸素種の1つとして認識されてきた。しかし,近年,過酸化水素が細胞の分化や成長,遊走に関わる情報伝達物質として生産されることが明らかとなり注目を集めている。例えば,上皮細胞の場合,上皮成長因子(EGF)などの刺激によるEGF受容体の自己リン酸化を契機として,膜タンパク質であるNADHオキシダーゼ(Nox)が活性化され,Noxによる酸素分子の還元により一旦,細胞外に過酸化水素が生産され,水チャネル蛋白質アクアポリンを通り,過酸化水素が細胞内に流入するとされている。昨年度までに,上記の情報伝達物質として生産される過酸化水素を細胞内で検出できる高感度な金属錯体型蛍光プローブMBFh1およびMBFh2を開発している。MBFh1およびMBFh2は,過酸化水素と反応し、秒単位で赤色蛍光物質レゾルフィンを放出する高感度な過酸化水素蛍光プローブである。本年度は、細胞の異なる領域での過酸化水素の発生(例えば、細胞の内外)をマルチカラーで捉えるために,過酸化水素との反応により緑色蛍光物質を生成する複数の金属錯体型蛍光プローブの合成を行った。その結果,ジヒドロジクロロフルオレセインと単核鉄錯体を有するプローブが良好な過酸化水素蛍光プローブとして機能し,細胞内の過酸化水素の検出に利用できることを見いだした(Y.
