Research Abstract

幹細胞の未分化状態での保持、培養並びに適切な組織細胞への分化誘導は、幹細胞を用いる次世代の医療技術である再生医療に必須である。幹細胞の運命の制御は、幹細胞培養基板の物理的特性(剛性/弾性)並びに生物化学的特性(細胞接着部位ペプチドの種類並びに表面密度)に依存するが、これらの研究は個別に行なわれてきており、系統的な研究はなされてこなかった。本研究では、様々な剛性/弾性を有する基板を作成し、その基板上に細胞接着因子ペプチドあるいは細胞外マトリックス蛋白質(ECM)を固定化させた基板を調製した。これら固定化細胞培養基板上において、ES 細胞、iPS 細胞、造血幹細胞並びに間葉系幹細胞を培養して、未分化状態を維持する最適な基板、あるいは、適切な組織細胞(骨芽細胞、インスリン産生細胞等)への分化効率の高い最適な基板の剛性/弾性並びに最適な細胞接着因子ペプチドあるいはECM を判明させることを本研究の目的とした。
ポリビニルアルコール・イタコン酸(PVA-IA)架橋ゲルを調製した。様々な剛性 /弾性(stiffness)を有する細胞培養基板の調製は、架橋反応時間(15分、30分、1時間、2時間、4時間、8時間、12時間、24時間48時間)を制御することにより行う。調製されたPVA-IA細胞培養基板の剛性 /弾性は、原子間力顕微鏡を用いて計測した。
造血幹細胞を架橋ゲル上で培養を行ったところ、フィブロネクチン細胞結合部位であるCS-1を固定化させた比較的弾性の低いゲルにおいて、造血幹細胞の増殖が増大することを見出した。本研究の成果より、造血幹細胞増殖における培養基板の最適な弾性率を始めて世界に報告した。

Reason

(1)様々な弾性率を有し、かつ様々な細胞培養ドメイン(細胞外マトリックス、細胞結合部位オリゴペプチド)を表面修飾した細胞培養基板の開発を行うことができた。(2)造血幹細胞の増殖に最適な弾性率を有し、最適なナノセグメントを有するバイオマテリアルを開発することができた。(3)幹細胞の分化に最適な基板設計における物理因子の効果をまとめて、総説として発表することができた(A. Higuchi Q.-D. Strategy for Future Research Activity

今後、(a) 様々な剛性 /弾性を有し、かつECM並びに細胞接着因子ペプチドが固定化された細胞培養基板の調製を行なう。細胞培養基板の剛性 /弾性は、原子間力顕微鏡により計測する。さらに、ナノセグメント固定化細胞培養基板上におけるiPS細胞並びにES細胞の培養を行う。
すなわち、ナノセグメント(ECM並びに細胞接着因子ペプチド)固定化細胞培養基板上でヒトES細胞(H9)並びにヒトiPS細胞(AH-1, 申請者が独自に脂肪由来幹細胞にretrovirusを用いてOct4, Sox2, c-Myc, Klf4遺伝子を注入して作成したセルライン化されたiPS細胞)をXeno-フリー培地[X-VIVO10培地(Lonza Walkersville製)+成長因子]中で培養する。7日おきに新しい細胞培養基板上に細胞の継体を行う。培地交換は毎日行い、半分の培地を新培地と交換する。5継体ごとに、ヒトES細胞(H9)並びにヒトiPS細胞(AH-1)の多分化能表面マーカー(Oct-3/4, Sox2, Nanog, SSEA-3, SSEA-4, TRA-1-81, TRA-1-73)の発現性を免疫染色法並びにフローサイトメトリー法を用いて確認する。また、5継体ごとに、テラトーマ形成能を評価するとともに、三胚葉細胞への分化能力を確認する。いかなる剛性/弾性を有する細胞培養基板がヒトES細胞(H9)並びにヒトiPS細胞(AH-1)の培養に適しているか(多分化能を長期的に維持しているか)、さらには、いかなるECM並びに細胞接着因子ペプチドがヒトES細胞(H9)並びにヒトiPS細胞(AH-1)の培養に適しているかを評価する。
